Decreased IgE Antibody Formation in Mice treated with Polyadenylic-Polyuridylic Acid and Polyinosinic-Polycytidylic Acid




Abstract
Effects of poly(A) * poly(U) or poly(I) * poly(C) were examined on the inhibition of IgE antibody response against Ascaris antigen in mice and the ability of their spleen cells to produce IFN-r and IL-4. When polt(A) * poly(U) or poly(I) *
Poly(C)
was
injected into the mice with the antigen simultaneously, suppression of IgE antibody formation was observed in poly(I) * poly(C)-treated mine, but not in poly(A) * poly(U)-treated ones. In contrast, when treatments of these adjuvants were done at
7
days
after immunization, decreased IgE antibody formations were observed in poly(A) * poly(U) or poly(I) * poly(C)-treated mice. But the inhibitory effect of poly(I) * poly(C) was significantly greater than that of poly(A) * poly(U). In order to
investigate
the effect of these adjuvants on IFN-r and IL-4 production by lymphocytes, spleen cells from poly(A) * poly(U) or poly(I) * poly(C)-treated mice, immunized with the antigen or not, were cultured with the same antigen or Con A for 48 hns.
Increased
IFN-r
production was found in the culture fluids of spleen cells from poly(A) * poly(I)-treated miice but not in poly(I) * poly(C)-treated mice but not in poly(A) * poly(U)-treated ones. In conclusion These data revealed that IFN-inducers, poly(A) *
poly(U)
and poly(I) * poly(C) suppress the IgF production of mice. However, their functional effects on the IFN-r and IL-4 production by spleen cells differ each other, suggesting that the decreased IgE production by these adjuvants may be mediated by
other
cytokines.
